Principles
The Canine E.canis Ab Test was detected by sandwich method and side flow immunohistochemistry.
The clinical symptoms of Canine Ehrlichiosis
The clinical symptoms of erich body infection are variable. Depending on the strain, the dog's immune response and infection with other tick - or flea-borne pathogens. It is important to keep in mind that some dogs may not show clinical or laboratory symptoms associated with ehrlichia infection, and some dogs may show severe signs. In general, ehrlichia seems to have more severe clinical symptoms than anaplasmosis. Clinical symptoms of Ehrlichia come in many forms and include nonspecific symptoms such as fever, fatigue, lethargy, anorexia, superficial lymphadenopathy
splenomegaly, hepatomegaly, or weight loss. Other signs have also been described, including vomiting, diarrhea, pain, movement intolerance, edema (hind limbs, tail, or scrotum), cough and/or dyspnea (with pneumonia), serous or mucopurulent eye and nasal discharge, miscarriage or neonatal death, and skin ulcers. Due to multiple arthritis, some studies have included claudication but there is no evidence to support this. Therefore, in the case of dogs infected with Ehrlichia, claudication and/or multiple arthritis, it is advisable to exclude co-infection with other pathogens. Common clinical symptoms of ehrlichia include pallor of the mucosa, prolonged bleeding during estrus due to anemia, nosebleed, petechiae, or ecchymosis, thrombocytopenia, thrombocytopenia-associated hematuria or melena, or vasculitis. The most common are anterior uveitis, corneal opacities, anterior atrial haemorrhage, retinal vascular tortuosity, retinal choroid lesions, subretinal hemorrhage, retinal detachment or blindness. Neurological symptoms are less common (typically secondary meningitis).
Test procedures of visual interpretation
1. Remove the test card from the foil pouch and place it flat on a work surface.
2. Transfer 1 drop (using a capillary dropper) of the treated sample into the sample well.
3. Transfer 3 drops (using a disposable dropper) of the assay buffer into the sample well.
4. Start the timer. Wait for 5 to 10 minutes to read the result.
Interpretation of visual interpretation results
Test procedures of instrumental interpretation (Standard Test)
1. Turn on the FIC-Q100N Dry Immunoassay Analyzer.
2. Use the analyzer's scanner to scan the QR code on the inner label of the box or the label of bag to import the calibration curve into the analyzer.
3. Remove the test card from the foil pouch.
4. Position the front of the card facing up, ensuring that the sample well is oriented towards the outside of the analyzer's test card slot.
5. Insert the card into the test card slot of the analyzer.
6. Use a pipette with a range of 2-20 μL to place 10 μL of the prepared sample into the sample well. Then, use a pipette with a range of 10-100 μL to place 90 μL of the assay buffer into the sample well.
7. Push the test card to the farthest end of the test card slot.
8. Tap "Standard Test" on the screen to start the testing. Please refer to the analyzer's operation manual for detailed instructions.
Test procedures of instrumental interpretation (Quick Test)
1. Turn on the FIC-Q100N Dry Immunoassay Analyzer .
2. Use the analyzer's scanner to scan the QR code on the inner label of the box or the label of bag to import the calibration curve into the analyzer.
3. Remove the test card from the foil pouch.
4. Transfer 1 drop (using a capillary dropper) of the treated sample into the sample well.
5. Transfer 3 drops (using a disposable dropper) of the assay buffer into the sample well.
6. Wait for 5-10 minutes, then push the test card to the farthest end of the test card slot.
7. Tap "Quick Test" on the screen to start the testing. Please refer to the analyzer's operation manual for detailed instructions.
Interpretation of instrumental interpretation results
With the assistance of the FIC-Q100N Dry Immunoassay Analyzer , the test result can be classified into five phases: negative, suspicious (grey zone), weak positive, positive, and strong positive. This method provides better judgment and more objective interpretation than visual judgment, especially in cases of low antibody titer. The symbols in the following table represent the different results and their interpretations.
