Escherichia coli K99 Antigen Rapid Test
Short Introduction
Enterotoxigenic Escherichia coli (ETEC) is an important pathogen that causes diarrhea in human and young animals (newborn piglets, calves, lambs and weaned piglets). After being infected with ETEC, newborn animals often die from severe watery diarrhea and rapid dehydration. Morbidity and mortality are high.
The Escherichia coli K99 Antigen Rapid Test is a lateral flow immunochromatographic assay for the qualitative detection of Escherichia coli K99 Antigen (E.coli K99 Ag) in animal's feces specimen.
Principle
The Escherichia coli K99 Antigen Rapid Test is based on sandwich lateral flow immunochromatographic assay. The test device has a testing window for the observation of assay running and result reading. The testing window has an invisible T (test) zone and a C (control) zone before running the assay. When the treated sample was applied into the sample hole on the device, the liquid will laterally flow through the surface of the test strip and react with the pre-coated Escherichia coli K99 antibodies. If there are anti-Escherichia coli K99 Antigen in the specimen, a visible T line will appears. The C line should always appear after a sample is applied, which indicates a valid result. By this means, the device can accurately indicate the presence of Escherichia coli K99 Antigen in the specimen.
Test Procedure
-Collect cattle's fresh whole blood, and get the serum or plasma specimen for use. Immediately apply the testing once collected the specimen.
-Allow all materials, including specimen and test device, recover to 15-25ºC before running the assay.
-Take out the test device from the foil pouch and place it horizontally.
-Using the disposable dropper to place 1 drop (approx. 40μL) of the prepared specimen into the sample hole "S" of the test device. Then drop 2 drops (approx. 80μL) of the assay buffer into the sample hole immediately.
-Interpret the result in 5-10 minutes. Result after 15 minutes is considered as invalid.
Interpretation Of Result
Positive: The presence of both "C" line and zone "T" line, no matter T line is clear or vague.
Negative: Only clear C band appear. No T band.
Invalid: No colored band appears in C zone. No matter if T band appears.
