African Swine Fever Virus Antibody Rapid Test
Short Introduction
African swine fever virus (ASFV) causes a severe disease in domestic pigs and wild boar that can result in death in almost all pigs that are infected. The clinical signs of ASF can vary but are similar to some other pig diseases. Signs typically occur 3-15 days after infection.
Clinical Signs
The early signs are non-specific:
High fever, lethargy, and loss of appetite.
Pigs may die suddenly without further disease signs.
At later stages:
Reddening of the skin (visible only in pale-skinned pigs), with patches appearing on the tips of ears, tail, feet, chest, or under the belly.
Diarrhoea, vomiting.
Laboured breathing.
Swollen red eyes, eye discharge.
Abortions, still-births.
Increasing morbidity and unwillingness to get up.
In severe cases death can sometimes be the only sign of infection, with a case fatality rate as high as 100%.
Principle
The African Swine Fever Virus Antibody Rapid Test is based on sandwich lateral flow immunochromatographic assay. The test device has a testing window for the observation of assay running and result reading. The testing window has an invisible T (test) zone and a C (control) zone before running the assay. When the treated sample was applied into the sample hole on the device, the liquid will laterally flow through the surface of the test strip and react with the pre-coated ASFV antigens. If there are anti-ASFV antibodies in the specimen, a visible T line will appears. The C line should always appear after a sample is applied, which indicates a valid result. By this means, the device can indicate the presence of African Swine Fever Virus antibodies in the specimen.
Test Procedure
- Collect pig's fresh whole blood, and get the serum or plasma specimen for use. Immediately apply the testing once collected the specimen.
- Take out the test device from the foil pouch and place it horizontally.
- Place 10μL of serum or plasma specimen into the sample well "S" on the test device. Then drop 2 drops of assay buffer into the sample well. Start the timer.
- Interpret the result at 10 minutes. Result after 15 minutes is considered as invalid.
Interpretation Of Result
- Positive (+): The presence of both "C" line and zone "T" line, no matter T line is clear or vague.
- Negative (-): Only clear C line appear. No T line.
- Invalid: No colored line appears in C zone. No matter if T line appears.
